Abstract

FORMULATION AND EVALUATION OF STOMACH SPECIFIC IN SITU GEL OF OFLOXACIN USING NATURAL, SYNTHETIC POLYMER

ABSTRACT

The aim of this study was to develop a novel gastro retentive oral floating in situ gel system for sustained drug delivery using natural biodegradable polymers. The formulations are designed with an objective to retain in stomach for an extended time period to obtain better bioavailability. Polymer based floating in situ gelling systems of Ofloxacin were prepared by dissolving varying concentrations of guar gum, methyl cellulose and sodium alginate in distilled water to which varying concentrations of drug and calcium carbonate are added . Sodium alginate was used as one of the bio-degradable polymer, calcium carbonate was used as cross-linking agent and guar gum act as a release retardant. The prepared formulations were evaluated for solution drug content, viscosity, sol to gel time, floating lag time, floating duration, gel strength, water uptake and in vitro drug release up to 12 hrs. The in vitro drug release was carried out using USP paddle type dissolution test apparatus in 0.1(N) HCl medium and it was found that the GG12 shows the best cumulative drug release upto 12 hrs of the period. From set of experiments, it was evident that guar gum plays a vital role in not only producing viscous solution but also controlling the release for longer duration that of sodium alginate. The in situ gel exhibited the expected viscosity, drug content and sustained drug release.
