Abstract

FORMULATION AND EVALUATION OF REPAGLINIDE TRANSDERMAL FILM

ABSTRACT

Low bioavailability of drug through skin is one of the major obstacles to the development of Transdermal drug delivery products. Several enhancers have been studied for permeation enhancer effect. Majority of them were found to produce undesirable effect on skin or body. So, to overcome this problem, we can use different quantities of plasticizers to optimize the best permeation of drug through the skin. The aim of present research was to develop matrix type transdermal film of repaglinide containing HPMC as polymer using solvent evaporation technique. Nine formulations containing different concentrations of three plasticizers (20% w/w, 30% w/w and 40% w/w of different polymer compositions namely propylene glycol, diethyl phthalate, dibutyl phthalate) were developed and evaluated for ex-vivo drug release through hairless rat skin. The ex-vivo permeation of drug through hairless rat skin was found to increase with the help of plasticizer. Formulation containing 20 % w/w of dibutyl phthalate was found to be best with respect to percent drug release in 24 hrs and shown the linearity in transdermal permeation.
