Abstract

CYTOTOXIC EFFECT OF REACTIVE VIOLET - 2RL DYE EXPOSED TO HUMAN EMBRYONIC KIDNEY CELL LINE (HEK293) BY MTT ASSAY

ABSTRACT

Textile industries are predominant in production and usage of dyes. Dyes are ease to synthesize and stronghold in fabrication. Textile effluents comprised of noxious substances has to be removed from the effluent run-off prior to discharge into the aquatic environment. Fungi have diverse application in degradation of dyes and idiosyncratic features like adsorption, degradation, decolorization and enzymatic degradation of dyes. Synthetic dye, Reactive Violet - 2RL was treated with Penicillium purpurogenum with varying concentrations (20 - 100 mg/l), the crude extort was assessed with HPLC for the confirmation of degradation of painstaking dyes and checked the toxicity with Human embryonic Kidney cell line (HEK - 293) by MTT assay. After 48 hrs incubation, the medium with MTT was then flicked off and the formed formazan crystals were solubilized in DMSO and absorbance read at 570 nm using micro plate reader. The percentage cell viability was then calculated and found to be Reactive Violet - 2RL (99.27%) , Reactive Violet - 2RL with C-limited medium (94.85%) and decolorized extort (81.48%) when exposed to high concentration of dye. The outcome of this research, demonstrates that there is drop-off in cell viability. This assessment will make possible to prevent the harmful effects and create an understanding in relation to the treatment technology.
