ABSTRACT

The study was focused to develop and validate a HPTLC method for estimation of Lafutidine from their pharmaceutical dosage form. The solvent system consisted of n-Butanol: Toluene: Chloroform: Ammonia (3:1:1:0.3 v/v/v/v). Densitometric analysis of Lafutidine was carried in the absorbance mode at 220 nm. This system was found to give compact spots for Lafutidine (Rf value of 0.52 0.02). The calibration curve of Lafutidine was found linear in the range of 1400- 2600 ng/spot with significantly high value of correlation coefficient. The linear regression analysis showed good linear relationship with r2 = 0.9996. The regression equation was found to be y= 4.0346x + 37.429. The developed method was validated for linearity, accuracy, system precision, method precision and recovery according to ICH guidelines. The method was found sensitive, accurate, precise and reproducible for determination of Lafutidine in tablet formulations and can be applied for the routine analysis of Lafutidine in marketed dosage forms.
