Abstract

PRODUCTION OF RIFAMYCIN SV USING IMMOBILIZED CELLS OF AMYCOLATOPSIS MEDITERRANEI - OVA5

ABSTRACT

Rifamycin is currently used as a basic molecule or intermediate for the synthesis of semi-synthetic antibiotics like rifampicin, rifabutin and rifap entine, which are clinically used as therapeutic agents. The purpose of the present work describes the evaluation, development of suitable culture substrate and the optimization of process parameters for production of Rifamycin SV using OVA5 (mutant strain) of Amycolatopsis mediterranei 5008 (NCIM) through SmF. Sodium alginate was found effective and suitable for improving the productivity of rifamycin SV. Rifamycin SV production with immobilized Amycolatopsis mediterranei cells was better matric made of 2% alginate and 4% calcium chloride with 30 minutes curing time and maintained at pH 7.0 while the temperature was maintained at 30ÂșC for a period of 144hrs. The rifamycin SV production increased which yielded higher quantity of antibiotic than free cells cultured under similar conditions.
