Abstract

ROLE OF TESTOSTERONE & FINASTERIDE ON HIGH FAT DIETINDUCED OBESITY IN WISTAR RATS

ABSTRACT

Objectives Testosterone level is decreased in obesity and low level of testosterone negatively effects the parameters of obesity and testosterone supplement shows beneficial effects in obesity in previous reports. Finasteride, a testosterone metabolism inhibitor increases the level of testosterone may positively effects in prevention of obesity. In present study orlistat a standard drug for obesity was used as positive control. The present study was designed to investigate the, role of testosterone and finasteride on high fat diet induced obesity in rat. Methods Male Wistar albino rats (170-210 g) (n=6) were employed. Experimental obesity was induced by feeding high fat diet (HFD) for 10 weeks. Body weight, body mass index, Lee index, weight of epididymal, retroperitoneal and mesenteric fat depots and serum glucose, cholesterol, HDL, LDL, VLDL and TG were assessed as an index of obesity. Key findings HFD treatment caused significant increase in body weight, body mass index, Lee index; weights of different fat depots; serum glucose, cholesterol, TG, LDL, VLDL; feed intake (Kcal); and decrease in serum HDL and feed intake (g) in HFD control group. Testosterone 100mg/kg/day and 200mg/kg/day and Finasteride 0.5mg/kg/day significantly attenuated the effects of HFD treatment. The anti-obesity effects of these drugs were comparable to orlistat, a well reported pancreatic lipase inhibitor. Conclusions: It may be concluded that the treatment with testosterone and finasteride significantly prevented the HFD-induced obesity. This provides a rational pharmacological basis for clinical use of these drugs in obesity.
