Abstract

CARDIOPROTECTIVE POTENTIAL OF A HYDROETHANOLIC POLYHERBAL CRUDE EXTRACT ON ISOPROTERENOL INDUCED MYOCARDIAL IN WISTAR ALBINO RATS

ABSTRACT

Atherosclerosis is complex disease and the underlying cause of heart attack, stroke and peripheral vascular disease. It is the main cause of morbidity and mortality worldwide, characterized by an excessive inflammatory, fibro fatty, proliferative response to damage of the artery wall. The present study was designed to evaluate the cardioprotective role of Polyherbal extract on isoproterenol induced myocardial infarction in Wistar albino rats. The rats were divided into eight groups of six animals each. Group I served as a normal control, Group II rats were administered isoproterenol (20mg/g B.wt). Group III and IV were pretreated with PH extract (250 mg/kg B.wt, 500mg/kg B.wt, respectively) for a period of 30 days and received a subcutaneous injection of isoproterenol (20mg/kg, B.wt) at the end of experimental period for 2 consecutive days. Group V and VI were pretreated with Propranolol (10 mg/kg B.wt, 20mg/kg B.wt) for a period of 30 days and received a subcutaneous injection of isoproterenol (20mg/kg, B.wt) at the end of experimental period for 2 consecutive days. Group VII and VIII received PH extract of 250mg/kg B.wt and 500mg/kg B.wt for 30 days. After the experimental period, blood was collected, serum and tissue was separated and used for the estimation of cardiac marker enzymes such as CKMB, CPK, AST, ALT, LDH, TROPONIN-T,TROPONIN-I,HOMOCYSTEINE, MPO and Membrane bound enzymes such as Na+/K+ ATPase, Ca2+ ATPase, Mg2+ ATPase are studied to accesses the cardio protection. Total protein was estimated in both the serum and heart tissue. The PH crude extract produced a significant (p<0.05) increase in total protein (serum, tissue), cardiac marker enzymes in tissue, membrane bound enzymes and a significant (p<0.05) decrease in cardiac marker enzymes in serum. The PH crude extract showed a significant (p<0.05) improvement in the treated groups. The effect of oral administration of PH extract at the dose of 500mg/kg B.wt was more than 250mg/kg B.wt. similarly the standard drug propranolol treated group 20mg/kg B.wt also showed a better result. This result indicates that the PH crude extract ex-hibit the cardioprotective activity and the protective effect could attribute to its membrane stabilizing action and validates the traditional use of this PH extract in myocardial infracted animals.
