Abstract

“ASSESSMENT OF ALPHA AMYLASE INHIBITORY ACTIVITY OF STEM AND ROOT EXTRACTS OF CARISSA CARANDAS LINN.”

Dr. Meenakshi Fartyal* and Padma Kumar

ABSTRACT

In the discovery of new pharmaceuticals, natural products have historically played a pivotal role, especially those derived from higher plants. In present study, alkaloids, flavonoids, steroids, petroleum ether, methanolic and water extract from stem & root of Carissa carandas Linn. were extracted and tested for antidiabetic activity, with salivary alpha amylase and starch as substrate using chromogenic DNSA (2,4- Di nitro Salicylic Acid) method & Starch iodine method. All experiments were performed in 3 different sets each in triplicates. The data are expressed as mean ± SEM (standard error of the mean). The highest inhibition for stem was found in its methanolic extract at the concentration of 1.5 mg/ml, with percent inhibition 58.00±2.00% & an IC50 value of 1.292 g/ml while in case of root, highest inhibition was obtained at 1.5mg/ml of methanolic extract, with % inhibition 68.94±3.60% & IC50 value 1.140 g/ml and at 1.5 mg/ml of pet ether extract, with % inhibition 58.34±2.39% & an IC50 value 1.201 g/ml. All the tested stem & root extracts have shown good inhibitory potential. Thus, stem & root extracts may be useful in managing the postprandial hyperglycemia and type2 diabetes either alone or in a combinatorial therapy.

Keywords: Antidiabetic activity, Alkaloid, Flavonoid, Steroid, Polar & non polar solvents, Salivary alpha amylase.
