FORMULATION AND EVALUATION OF BUCCAL TABLETS OF RIZATRIPTAN BENZOATE

Abstract The study aim was concerned with formulation and evaluation of bioadhesive buccal drug delivery of Rizatriptan Benzoate tablets, which is extensively metabolized by liver. The tablets were prepared by direct compression using bioadhesive polymers such as hydroxypropyl methylcellulose K15M, hydroxypropyl methylcellulose K100M sodium carboxymethyl cellulose and Carbopol 971 P. Bioadhesion strength, ex vivo residence time, swelling, and in vitro dissolution studies and ex vivo permeation studies were performed. Trial batches were taken with all four polymers and finally 32 full factorial design was applied by taking HPMC K15M and Carbopol 971 P as independent variables. The effect of independent variables was checked on Drug release at 1 hour and mucoadhesive strength of all factorial batches. The Quadratic model found significant and both factors gave significant effect in responses proved by ANOVA analysis. Final batch was taken from the overlay plot after validating the design and the formulation found stable up 1 month, gives sustained release dissolution profile up to 8 hours and good mucoadhesive strength.
