EVALUATION OF CARDIOPROTECTIVE AND ANTIOXIDANT ACTIVITY OF NOTHAPODYTES NIMMONIANA

Abstract Objective: To investigate the possible Cardioprotective and Antioxidant activity of the ethanolic extract of Nothapodytes nimmoniana (NN) on 5-Fluorouracil induced cardiotoxicity in rats. Experimental design: The Nothapodytes nimmoniana stem powder were preparation of ethanolic extract followed by evaporation to dryness. For cardioprotective and Antioxidant activity, 30 rats were divided into 5 groups. Group-1 as normal control, group-2 as disease control, group- 3 as standard (Vit-E), group-4 as test drug low dose and group-5 as test drug high dose. The treatment is given for 7 days and on 5th day, 5-Fluorouracil (150mg/kg i.p.) was administered to induce cardiac toxicity. NN 200 and 400 mg/kg I.P. was given to all the groups for 7 days. Fortyeight hours after the 5-Fluorouracil administration, all the rats were killed in order to withdraw the blood for the examination of Creatine kinase-MB (CK-MB), aspartate transaminase (SGOT), alanine transaminase (SGPT), total cholesterol (TC), LDL and HDL. Results and Conclusion: Rats treated with ethanolic extract of Nothapodytes nimmoniana (200 mg/kg and 400 mg/kg) showed significant decreased in CK-MB, SGOT, SGPT, TC, LDL and increases HDL levels. The higher dose of Nothapodytes nimmoniana 400 mg/kg showed more significant cardioprotection compared to lower dose 200 mg/kg. The antioxidant studies of Nothapodytes nimmoniana with high dose test group (400 mg/kg) showed significant increase in the activities of SOD, CAT and GPO and significant decrease in LPO compared with disease control group. The histopathological studies showed the plant extract significantly minimized the cardiotoxicity induced by 5- fluorouracil.
