Abstract

EFFECT OF CDRI 85/287 ON HISTOCHEMICAL LOCALIZATION OF UTERINE KEY ENZYMES IN RAT DURING PRE-IMPLANTATION PERIOD

Abstract

The present study was undertaken to determine the effect of a potent oestrogen antagonist and antiimplantation agent, CDRI 85/287 on histochemical localization of certain key enzymes involved in the preparation of uterine endometrium for receptivity of the blastocyst and consequently the process of implantation. The enzymes measured involved ß-glucuronidase, peroxidase, leucine-aminopeptidase (LAP) and alkaline phosphatase (ALP). Results revealed differential cellular distribution pattern of all enzymes in the uterine tissue. While the uterine peroxidase and ALP enzyme activity showed increasing trend from day 3-5 post-coitum (p.c.). On the other hand ß-glucuronidase and LAP activity increased from day 3-4, with maximam activity on day 4 p.c. On day 5 p.c. the decline was noticed in the staining intensity of these enzymes in normal pregnant rats. Administration of 85/287 on day 1 p.c. at its contraceptive dose (2.5 mg/Kg, p.o.) caused inhibition in uterine peroxidase activity (on days 4 and 5) and ALP activity both in stroma and epithelium except blood vessels which were strongly stained. The activity of ß-glucuronidase and LAP exhibited a decrease on day 4 as compared to control rat uterus. However, on day 5 p.c. LAP activity showed a decrease in the staining intensity in treated rats. Results of this study indicate hormone dependent modulations in the localization of these enzymes on different days of pre-implantation period/implantation day there by suggesting inhibition of estrogenic action required for preparation of uterine endometrium for receptivity of the blastocyst/attachment reaction under the influence of CDRI-85/287. The results are discussed in relation to the inhibitory activity of this novel molecule in implantation.
