SYNTHESIS, CHARACTERIZATION AND BIOLOGICAL ACTIVITY OF NOVEL 1,3,4-OXADIAZOLE DERIVATIVES

Abstract Various substituted 1,3,4-oxadiazole derivatives have received considerable importance during last decade as they are covered with wide verity of biological activities and have a wide range of therapeutic importance. Based on this a series of new 1,3,4-oxadiazole derivatives have been synthesized. Three novel 1,3,4-oxadiazole derivatives were synthesized by simple facile method. salicylic acid upon reacts with hydrazine hydrate to form benzohydrazide. The benzohydrazide upon treat with Pocl3 and PABA to form oxadiazole. The formed compound upon treatment with substituted aromatic aldehydes in the presence of concentrated HCl and ethanol with 2 hours reflux finally to give Schiff bases like ODZ-1 to ODZ-3. The synthesized compounds were characterized by physical studies like solubility, melting point, TLC and subjected to spectral studies like IR,1H-NMR and mass spectroscopy. All the synthesized compounds were screened for invitro anti-tubercular activity was performed by microplate alamar blue assay method. Among three novel 1,3,4- oxadiazole ODZ-3 derivative shows significant activity when compared with standard drugs (ciprofloxacin, pyrazinamide and streptomycin).
