DEVELOPMENT AND EVALUATION OF GASTRORETENTIVE RAFT FORMING TABLET OF HYDROCHLOROTHIAZIDE

Abstract In the present study an attempt was made to formulate Gastroretentive Raft forming tablet of Hydrochlorothiazide which was used in the treatment of Diuresis. In this system drug remained buoyant in gastric region for several hours in order to prolong its gastric residence time and maintain constant therapeutic level over prolonged period of time. Guar gum, Chitosan, HPMC K100M were the polymers used in different concentration ratios in comparison to Sodium Alginate to formulate best suitable formulation. Tablets were prepared by direct compression method and subjected to various evaluation parameters which include Raft strength measurement, Raft weight and Raft volume determination, in-vitro drug release, etc. DSC thermographs showed that the drug is compatible with polymer and excipients. Formulation containing 100 mg of Sodium Alginate and 25 mg of Chitosan showed highest drug release of 92.68±0.71%, good Raft strength, good Raft weight, better Raft volume and thus formulation F4 was considered as optimized. Formulated gastroretentive raft forming tablet of Hydrochlorothiazide seems to be safe and an effective approach for oral controlled drug delivery.
