ENHANCING BIOAVAILABILITY OF GLIBENCLAMIDE BY THE PREPARATION AND EVALUATION OF FAST DISSOLVING NANO FIBER PATCH

Abstract In present investigation we have designed nanofiber patch of Glibenclamide to increase the retention time, bioavailability, onset of action and reducing the pre systemic metabolism. This will ultimately deliver an optimum therapeutic concentration of Glibenclamide in systemic circulation to exert its pharmacological action. Electrospun nanofibers have received special attention as drug delivery systems because of their distinct functional feature and easy fabrication techniques. Glibenclmide is a second-generation sulfonylurea antidiabetic agent. Glibenclamide bind to ATP-sensitive potassium channels on the pancreatic cell surface, reducing potassium conductance and causing depolarization of the membrane. Depolarization stimulates calcium ion influx through voltage-sensitive calcium channels, raising intracellular concentrations of calcium ions, which induces the secretion, or exocytosis, of insulin.
