DETERMINATION OF FIBRINOLYTIC ACTIVITY OF ETHANOIC EXTRACT OF LAWSONIA INERMIS (HENNA) SEEDS IN VITRO STUDY

Abstract Objectives: Lawsonia inermis (henna) seeds is a natural product harvested from plant fruit which is grow in different climate. The present study aimed to determine the fibrinolytic effect of ethanolic extract of lawsonia inermis (henna) seeds on whole blood (WB) clot through an in vitro study. Methods: Standardized WB clots were prepared and incubated in platelet poor plasma (PPP) as control and in 1.0 mg/ml (diluted in PPP) for different incubation time: 3, 6 and 9 hours. The plasma from these tubes was measured individually for Ddimer (DD) using immuno-turbidometric method and residual of WB clots were weighed. Repeated measures were done. ANOVA statistical test was performed for data analysis. Results: There was significant mean difference of DD between PPP and lawsonia inermis extract groups based on time (P<0.001). The mean DD was higher in lawsonia inermis extract group compared to PPP (normal control) groups for all the incubation times (3, 6 and 9 hrs). The mean DD (SD) from PPP (control) recorded at 3, 6 and 9 hrs were 1.02 (0.15), 1.25 (0.12) and 1.37 (0.09) 𝜇g/ml respectively. The mean DD (SD) from plasma containing lawsonia inermis extract at 3, 6 and 9hrs were: 1.33(0.27), 2.05 (0.23) and 3.6 (0.61) μg/ml respectively. There was a significant difference of median (IqR) of WB clot weight between pre and post incubation in PPP (normal control) and in lawsonia inermis extract at 3 hrs, 6 hrs and 9 hrs (p-values <0.01). However there was insignificant different of clot weight between normal control and lawsonia inermis extract treated whole blood. Conclusion: lawsonia inermis extract at 1.0 mg/ml concentration demonstrated fibrinolytic activity based on DD quantitation and not by clot weight by in vitro WB clot lysis in this study. Lawsonia inermis showed time dependent activity and a potential antithrombotic agent requiring further investigation. Keywords: Lawsonia inermis, Fibrinolysis, D-Dimer and Whole blood clot.
