Abstract

FORMULATION, CHARACTERIZATION AND DRUG RELEASE CHARACTERISTICS OF DEXTRAN-PLGA-BASED NANOPARTICLES LOADED WITH TAMOXIFEN CITRATE

Abstract

Tamoxifen Citrate is a highly potent drug in for treatment of cancer. However, lower water solubility and dose-dependent side effects like neutropenia, peripheral neuropathy and hypersensitivity reaction limit it’s clinical applications. To improve the delivery aspects of tamoxifen citrate, we have fabricated the dextran-PLGA- based polymeric micelles of the drug. First of all, dextran-PLGA was prepared and characterized by IR and NMR analysis. Micelles were prepared via precipitation dialysis method. CMC was determined I/KI probe method. Particle size and zeta potential were determined Delsa Nano Particle Analyser. Particle size was found less than 100 nm. Polymeric micelles fabricated dextran-PLGA block copolymer was spherical and smaller in size (in nanometer range). In vitro drug release was performed in phosphate buffer saline (pH 5.8) and it exhibirted the controlled release of drug. Cell cytotoxicity study showed that the tumor cells (MCF-7 breast cancer cell line) cytoxicity by the tamoxifen citrate was higher as compared to free tamoxifen citrate. Higher engulfment in tumor cells indicates specific targeting and more uptake of polymeric micelles.
