. Abstract Objectives: The present study was carried out with an aim of evaluating anti-Parkinson’s activity of a polyherbal extract (PHE) containing Prunus dulcis, Nigella sativa, Cucurbita pepo, Piper nigrum and Piper longum, in reserpine and haloperidol-induced Parkinson’s Disease (PD) models. Methodology: Haloperidol and Reserpine were used to induce PD in healthy male Swiss albino rats. PHE was given in three different doses (100mg/kg, 200mg/kg and 400mg/kg). Behavioral and locomotor activities of PHE-treated groups were assessed and equated with the standard drug-treated group. Results: Administration of haloperidol and reserpine caused decreased behavioral and locomotor activities in rats which were later restored by administration of PHE. PHE therapy causes considerable changes in locomotor activity in PD-affected mice, indicating that it has anti-Parkinson's potential. Conclusion: The anti-Parkinson’s activity of PHE was found to be dose-dependent with 400mg/kg being the most suitable dose for its optimum activity.
