MECHANISTIC EVALUATION OF TRICHOSANTHES DIOICA WALL. PLANT ON MOUSE GLIOMA CELLS

. Abstract We evaluated the antiproliferative activity of methanolic extracts of Trichosanthes dioica Wall. leaves (METDL) and root (METDR) on mouse glioma cells. Both the extracts and their active fraction significantly inhibited cell viability in a time and concentration dependent manner. Morphological study showed several signs of apoptosis and degraded DNA bands of glioma cells with both the treated cells than control. Treated cells have exerted significant MMP shift in the mitochondrial transmembrane potential activity involving the release of cytochrome C into the cytosol. The presence of apoptotic cells in the early and late stages was confirmed by the flow cytometric analysis. Cell cycle phase arrest was observed in the G0/G1 phase in the METDL and METDR treated cells. Caspase assay reveals that the pathway followed may be intrinsic apoptosis pathway. These findings suggest that both the extracts possess anti-glioma effects via apoptosis not necrosis. Keywords: Trichosanthes dioica Plant, Cancer, Glioma, Apoptosis, Caspase.
