IMPROVING BLOOD GLUCOSE REGULATION ON ALLOXAN MONOHYDRATE-INDUCED DIABETIC MICE: A SYNERGISTIC EFFECT OF PIPERINE AND LINAGLIPTIN

. Abstract Aim: High blood sugar levels are the outcome of diabetes mellitus, a metabolic and chronic condition caused by insufficient insulin secretion, action, or both. The main purpose of this study is to evaluate the effect of piperine with linagliptin on the lowering of blood glucose levels in alloxan- induced diabetic mice through the estimation of blood glucose levels and the mean plasma concentration of linagliptin. Methods: Based on randomization technique, Alloxaninduced( 150mg/kg,i.p.) thirty two CD-1 Male and Female Mice (aged 8-9 weeks old)were divided into 4 groups, Group 1(Disease control)can be administered with 0.5% CMC, Group 2(Drug alone) animals of both sexes male and female were administered with Linagliptin(1mg/kg,p.o.) and Group 3(Therapeutic dose) in this all the animals were administered with both piperine and linagliptin (10mg/kg, p.o.+1mg/kg,p.o.) and Group 4(Sub-therapeutic dose) in this all the animals were administered with both piperine and linagliptin (10mg/kg, p.o.+0.3mg/kg, p.o.). Blood glucose levels were estimated at day 1,7 and 14. On the 14th day of treatment the animal were anaesthetized, blood samples were collected through the saphenous vein method and plasma were separated for the estimation of mean plasma concentration of linagliptin. Results: The findings of this study are that a combination of piperine with linagliptin therapeutic dose (10 mg/kg + 1 mg/kg,p.o.) showed significantly higher lowering of blood glucose levels as compared to linagliptin alone (1 mg/kg, p.o.) and also the Disease control groups on both the 7th and 14th days (p Keywords: Diabetes, Bio-enhancingeffect, piperine, Linagliptin, bioavailability, AUC, Cmax, tmax, LC-MS/MS, Blood glucose levels, mean plasma concentration of linagliptin.
