FORMULATION AND EVALUATION OF TRANSDERMAL PATCHES LOADED WITH AMIODARONE

Abstract

Objective: Amiodarone is class III anti arrhythmic drug with 35 to 65% bioavailability. Trandermal delivery of drugs is known to improve the bioavailability of poorly soluble drugs. Transdermal patch of amiodarone was intended in the investigation. Methods: In the present investigation we report the formulation and evaluation of transdermal patches loaded with amiodarone (AMD) using hydroxypropyl methyl cellulose (HPMC) and ethyl cellulose (EC) as the film forming polymeric combination, applying the solvent evaporation technique. Results: The thickness of these patches ranged from 0.11 to 0.19 mm and average weight from 351 to 407 mg. The folding endurance was more than 30 for all the batches. The amount of moisture absorption as well as loss was in the range of 10.35 to 12.22 and 5.17 to 7.99 % respectively. The tensile strength ranged from 10.12 to 12.39 Kg/cm2 suggesting sufficient strength in the patches. The drug content in the formulations ranged from 97.91 to 99.18%. The best patch formulation was found to be AP7 and exhibited good physicochemical as well as release of AMD. Conclusion: The controlled release of drug from the patches makes it very much suitable for sustained release of AMD in much lower doses thus improving the overall pharmacokinetic profile of the drug.
