ANTI-TUMOR, ANTI-INFLAMMATORY AND ANTI-OXIDANT EFFECTS OF WORMWOOD (ARTEMISIA MONOSPERMA A.) OIL EXTRACT ON THE RISKS OF COLON CANCER INDUCED IN RATS

Abstract Objective: This study was undertaken to investigate the anti-tumor, anti-inflammatory and anti-oxidant potentials of oil extracts of Artemisia monosperma A. (wormwood) in vivo against dinitrochlorobenzene (DNCB) for chemical induction of colorectal cancer (CRC). methods: Malignant rats were orally administered DNCB at dose 24×10-3g dissolved in 1.2 ×10-3 L acetone 95% per kg body weight (day after day) for 2 weeks then injected in colon with 1×10-3g dissolve in 1mL ethyl alcohol 50% per kg body weight for 3 weeks (once/week). Wormwood oil extract (WO) was administrated (600mg/kg/body weight) as a daily oral dose. Rats were divided into 6 groups; namely G1:(Healthy control), G2:(Healthy WO), G3:(Malignant control),G4:(WO Protection), G5:(WO Treatment) And G6:WO (Protection + Treatment). Results: The antitumor and anti- inflammatory potentials of WO was evident through the significant reduction in serumAlkaline phosphatase (ALP), tumor necrosis factor-α(TNF)-α, alpha-feto protein(AFP), carcinoembryonic antigen(CEA), cancer antigen 19.9(CA-19.9),C-reactive protein(CRP), erythrocyte sedimentation rate (ESR) and interleukin-6(IL-6). Furthermore, the oil extract had an antioxidant effect as reflected by elevated the depleted oxidative stress markers including TAC, GSH and CAT and SOD activities and also significantly reduced the marked elevation of MDA and NO levels. Moreover, WO significantly improved nutritional and hematological parameters in Malignant rats. Interestingly, the group G6 of the Protection + Treatment design exerted almost the most protection of CRC compared to the other treated groups (G4 and G5). Furthermore, these results were confirmed by histopathological examination of the colon. In conclusion: WO administration significantly Suppressed tumorgenesis and has anti-inflammatory and anti-oxidant effects.
